目的:研究局部应用重组人干扰素-a2b(rhIFN-a2b)软膏后有无透皮及其在皮肤内的浓度。方法:选用乳猪皮肤,体外固体制剂溶出度仪-旋转膜装置药物供给系统和体内给药系统,抗人IFN-a单克隆抗体ELISA检测。结果:离体试验各时间点接受液中均未测得rhIFN-a,而离体和体内试验皮肤匀浆中均能检测到rhIFN-a。结论:rhIFN-a2b软膏能透入皮肤内,其在皮肤内的浓度达到该药在体外抑制I、II型单纯疤疹病毒IC50值的1.3-4.3和3.3-9.9倍。
OBJECTIVE: To investigate the presence or absence of transdermal and intradermal concentrations of rhIFN-a2b topical ointment. Methods: Superoxide dismutase (HRP) - drug delivery system and in vivo drug delivery system of swine porcine skin were used to detect anti-human IFN-a monoclonal antibody. RESULTS: No rhIFN-a was detected in the fluid received at all time points in vitro, whereas rhIFN-a was detected in both in vitro and in vivo skin homogenates.
