以香菇柄为原料,采用超声-闪式联合方法提取麦角甾醇,探讨提取时间、闪提转速、液料比、超声功率对麦角甾醇提取的影响。在单因素试验的基础上,通过响应面法优化超声-闪式联合提取香菇柄中麦角甾醇的工艺参数,利用S180荷瘤小鼠模型和酶联免疫吸附试验(ELISA)研究麦角甾醇的抗肿瘤活性及其机制。结果表明,利用超声-闪式联合法提取香菇柄中麦角甾醇的优化工艺参数为:室温条件提取14 min,闪提转速10 000 r/min,液料比36 m L/g,超声功率450 W,此条件下香菇柄中麦角甾醇的提取率为2.818 mg/g。可见超声-闪式联合法提取时间短,提取率较高;麦角甾醇高【400 mg/(kg·d)】、中【300 mg/(kg·d)】、低【(200 mg/(kg·d)】剂量组的瘤重抑制率分别为41.64%,29.90%,19.52%,与模型对照组相比均有显著性差异(P<0.05),小鼠血管内皮生长因子(VEGF)A的表达水平也较模型组显著降低(P<0.05),说明香菇柄麦角甾醇具有抗肿瘤活性,其抑瘤机制可能与抑制VEGF-A生成有关。
Using the mushroom stem as raw material, the ergosterol was extracted by ultrasonic-flash combined method. The effects of extraction time, flash rotation speed, liquid to material ratio and ultrasonic power on ergosterol extraction were discussed. Based on the single factor experiment, the process parameters of ergosterol in the mushroom stem were optimized by response surface methodology and the anti-tumor effects of ergosterol were studied by S180 tumor-bearing mouse model and enzyme-linked immunosorbent assay (ELISA) Activity and mechanism. The results showed that the optimal parameters of extraction of ergosterol from the stems of mushrooms by ultrasonic - flash method were: extraction at room temperature for 14 min, flash speed of 10 000 r / min, liquid - to - liquid ratio of 36 m L / g, ultrasonic power of 450 W Under this condition, the extraction rate of ergosterol in the stem of mushroom was 2.818 mg / g. It can be seen that the extraction time is short and the extraction rate is high with ultrasonic-flash combined method; the high [400 mg / (kg · d)] ergosterol, [300 mg / (kg · d) The tumor weight inhibition rates of the dose group were 41.64%, 29.90% and 19.52%, respectively, which were significantly lower than those of the model control group (P <0.05). The inhibitory rates of vascular endothelial growth factor (P <0.05), indicating that mushroom mushroom ergosterol has anti-tumor activity, and its anti-tumor mechanism may be related to the inhibition of VEGF-A production.
