目的:观察抗柯萨奇B病毒性心肌炎胶囊(K-CoxB-JN)总黄酮对阿霉素所致小鼠急性心肌损伤的作用,并探讨其可能作用机制。方法:选取正常昆明种小鼠175只,随机分为7组,即K-CoxB-JN黄酮高、中、低剂量组(0.8,0.4,0.2 g.kg-1),模型组,心血康组,正常组,原方水煎组,每组25只,每天ig 1次,1周后ip阿霉素20 mg.kg-1建立小鼠急性心肌损伤模型,24 h后眼球取血测定血清心肌酶水平、超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量,测定心脾指数。结果:K-CoxB-JN总黄酮能显著降低小鼠心肌酶活性,黄酮中剂量组乳酸脱氢酶(LDH)水平有所下降,α-羟丁酸脱氢酶(HBDH)水平降低明显(P<0.01);黄酮高、中剂量组心脏指数明显降低(P<0.01),黄酮中剂量组脾指数有显著性差异(P<0.05)。黄酮高、中剂量组SOD活力明显升高(P<0.01),黄酮中剂量组MDA含量有显著性降低(p<0.01),以黄酮中剂量效果最为明显。结论:K-CoxB-JN总黄酮对小鼠急性心肌损伤有保护作用,其作用机制可能与抗氧化、清除氧自由基有关。
Objective: To observe the effect of total flavonoids of CoxB-JN against acute myocardial injury induced by doxorubicin in mice and to explore its possible mechanism. Methods: A total of 175 normal Kunming mice were randomly divided into 7 groups: high, medium and low dose K-CoxB-JN flavonoids (0.8,0.4,0.2 g.kg-1), model group, , The normal group, the original side decoction group, each group 25, ig 1 day, 1 week after ip doxorubicin 20 mg.kg-1 mouse acute myocardial injury model, 24 h after the eyeball blood serum Enzyme activity, superoxide dismutase (SOD) activity and malondialdehyde (MDA) content were measured. The indexes of heart in high and medium dose flavonoids group were significantly lower (P <0.01), while the spleen index of middle dose group was significantly different (P <0.05). The activities of SOD in high and medium dose flavonoids groups were significantly increased (P <0.01), and the content of MDA in middle flavonoid groups was significantly decreased (p <0.01). Conclusion: K-CoxB-JN flavonoids have a protective effect on acute myocardial injury in mice and its mechanism may be related to anti-oxidation and scavenging oxygen free radicals.
