目的研究甲氨蝶呤(MTX)对中药复方银屑灵2号主要成分芍药苷药代动力学参数的影响,评价联合用药的合理性。方法将12只SD大鼠随机分为银屑灵2号组、联合用药组(MTX+银屑灵2号),单次口服给药后在不同时间点眼眶静脉采血,固相萃取(SPE)处理,采用HPLC测定血浆中芍药苷浓度,观察芍药苷在两组中的药代动力学参数的变化。结果与银屑灵2号组比较,联合用药组芍药苷AUC_(0-t)增加了29.50%,C_(max)增加了35.32%,T_(max)延迟了0.067 h,清除率降低了41.99%。结论 MTX与银屑灵2号联用可增加银屑灵2号中芍药苷的血药浓度,降低清除率,提高生物利用度。
Objective To study the effect of methotrexate (MTX) on the pharmacokinetics parameters of paeoniflorin, the main component of Psoriasis No.2, to evaluate the rationality of combination therapy. Methods Twelve Sprague-Dawley rats were randomly divided into two groups: Psoriasis 2 group and combination group (MTX + Yinxianling No.2). After a single oral administration, orbital venous blood was collected at different time points, followed by solid phase extraction (SPE) , The concentration of paeoniflorin in plasma was determined by HPLC, and the changes of pharmacokinetic parameters of paeoniflorin in both groups were observed. Results Compared with Yin Dalai No.2 group, the combination group paeoniflorin AUC_ (0-t) increased 29.50%, C_ (max) increased 35.32%, T_ (max) delayed 0.067 h, clearance rate decreased by 41.99% . Conclusion Combination of MTX and Yinza Ling 2 can increase the concentration of paeoniflorin in Yinzailing No.2, reduce the clearance rate and improve the bioavailability.
