目的:探讨小干扰RNA对大鼠血管内皮细胞细胞间黏附分子-1(ICAM-1)表达的抑制作用。方法:使用ICAM-1siRNA及错配siRNA转染肿瘤坏死因子-α刺激过的大鼠血管内皮细胞,聚合酶链反应和蛋白印迹检测转染后48hICAM-1mRNA和蛋白表达水平。结果:与空白对照组比较,大鼠血管内皮细胞在转染48h后ICAM-1mRNA表达减少90.0%,蛋白表达减少83.0%。结论:使用小干扰RNA能有效抑制大鼠血管内皮细胞ICAM-1表达。
Objective: To investigate the inhibitory effect of small interfering RNA on the expression of intercellular adhesion molecule-1 (ICAM-1) in rat vascular endothelial cells. Methods: ICAM-1 siRNA and mismatched siRNA were used to transfect tumor necrosis factor-α-stimulated rat vascular endothelial cells. The expression of ICAM-1 mRNA and protein was detected 48 h after transfection by polymerase chain reaction and Western blotting. Results: Compared with the blank control group, the expression of ICAM-1 mRNA decreased by 90.0% and the protein expression decreased by 83.0% in rat vascular endothelial cells 48 h after transfection. Conclusion: The use of small interfering RNA can effectively inhibit ICAM-1 expression in rat vascular endothelial cells.
