目的建立辐照水产品中邻酪氨酸的高效液相色谱检测方法。方法样品经0.1%甲酸均质,超声促溶后,离心去除不溶物质,加丙酮沉淀多余蛋白后,挥发有机试剂,冻干后复溶,高效液相色谱检测,外标法定量。采用Aglient ZORBAX Eclipse-C18色谱柱,甲醇-0.1%甲酸为流动相,激发波长为275 nm,发射波长为305nm,流速为0.8 m L/min。结果样品在50、200、500μg/kg添加水平的平均回收率72.20%~85.68%,相对标准偏差(RSD)为1.716%~9.926%。结论该方法可用于电子束辐照水产品中邻酪氨酸含量的检测,不同种类水产品中游离酪氨酸含量由高到低依次为基围虾>鲈鱼>扇贝。
Objective To establish a method for the determination of o-tyrosine in irradiated aquatic products by high performance liquid chromatography. Methods The samples were homogenized with 0.1% formic acid, dissolved in water and then centrifuged to remove insoluble materials. After excess acetone was precipitated by acetone, the organic solvents were evaporated, lyophilized and reconstituted. The samples were determined by high performance liquid chromatography and quantified by external standard method. An Aglient ZORBAX Eclipse-C18 column was used with methanol-0.1% formic acid as the mobile phase at an excitation wavelength of 275 nm and an emission wavelength of 305 nm at a flow rate of 0.8 mL / min. Results The average recovery was 72.20% ~ 85.68% and the relative standard deviation (RSD) was 1.716% ~ 9.926% at the sample concentration of 50,200 and 500μg / kg. Conclusion The method can be used for the detection of ortho-tyrosine in aquatic products irradiated by electron beam. The contents of free tyrosine in different aquatic products are shrimp> perch> scallop in descending order.
