目的:观察头痛宁胶囊对偏头痛模型大鼠三叉神经节P2X3mRNA表达的影响。方法:健康SD大鼠随机分为:正常对照组(生理盐水组)、模型组(偏头痛模型组)、西比灵组1.04 mg.kg-1、头痛宁高、中、低剂量组(760,380,190 mg.kg-1)。预防给药7 d。第7天ig 30 min后sc硝酸甘油(NTG,10 mg.kg-1)制备偏头痛大鼠模型,造模后3,6 h 2个时间点取材,用RT-PCR方法检测各组大鼠三叉神经节P2X3mRNA的表达情况。结果:与正常对照组比较,模型组大鼠三叉神经节P2X3mRNA表达数增多(P<0.01),3 h多于6 h。与模型组比较,头痛宁高、中、低剂量治疗组和西比灵治疗组大鼠三叉神经节P2X3mRNA表达数均减少(均P<0.01)。结论:头痛宁胶囊可能通过抑制由硝酸甘油诱导的P2X3mRNA过度表达,发挥治疗偏头痛的作用。
Objective: To observe the effect of TATT on P2X3 mRNA expression in trigeminal ganglion of migraine model rats. Methods: Healthy SD rats were randomly divided into normal control group (normal saline group), model group (migraine model group), xipingling group (1.04 mg.kg-1), Totaining high, middle and low dose group mg.kg-1). Prevention of medication for 7 days. Migraine rat model was prepared by nitroglycerin (NTG, 10 mg.kg-1) 30 min after ig for 30 min on the 7th day. Results: Compared with the normal control group, the expression of P2X3 mRNA in the trigeminal ganglion increased (P <0.01) in the model group more than 6 h after 3 h. Compared with the model group, the expression of P2X3mRNA in the trigeminal ganglion of the headache Ning high, medium and low dose treatment groups and the Xibeiling treatment group were decreased (both P <0.01). Conclusion: TONGTTNING capsule may play a role in the treatment of migraine by inhibiting P2X3 mRNA overexpression induced by nitroglycerin.
