我们以反相高效液相色谱法(RP-HPLC)和放射免疫测定法(RIA)研究了粉防己碱(Tet)对体外用钙离子载体A_(23187)诱导兔多形核白细胞(PMN)合成白三烯B_4(LTB_4)和血栓素A_2(TXA_2)的影响,并以地塞米松(Dex)和氟奋乃静(FPZ)作对照。结果表明,Tet与PMN温育15min即可抑制PMN合成LTB_4和TXA_2,其效应呈剂量依赖性。加入外源性花生四烯酸(AA)时,Tet的上述抑制作用消失。FPZ与Tet有相似的作用,Dex则与Tet不同,它需与PMN温育60min才出现抑制作用。
We investigated the effect of tetrandrine on the synthesis of rabbit polymorphonuclear leukocytes (PMN) induced by calcium ionophore A 23187 in vitro using reverse-phase high-performance liquid chromatography (RP-HPLC) and radioimmunoassay (RIA) Leukotriene B 4 (LTB 4) and thromboxane A 2 (TXA 2). Dex and flunarizine (FPZ) were used as controls. The results showed that Tet and PMN could inhibit the synthesis of LTB_4 and TXA_2 by PMN in a dose-dependent manner. With the addition of exogenous arachidonic acid (AA), Tet above the disappearance of inhibition. FPZ and Tet have a similar effect, Dex and Tet is different, it needs to be incubated with PMN 60min before inhibition.
