目的 :探讨基质金属蛋白酶抑制剂 (TIMP 2 )对SGC 790 1胃癌细胞体外侵袭能力的影响。方法 :将人全长TIMP 2基因经脂质体包裹转染PA317细胞系 ,G418筛选 ,感染人SGC 790 1胃癌细胞系 ,经体外细胞增殖、细胞电泳率、软琼脂形成和侵袭小室的实验综合分析其生物学行为改变。结果 :经TIMP 2基因转染的SGC 790 1细胞和MFC细胞和细胞电泳率。软琼脂形成和侵袭能力均明显低于对照组 ,而与对照组相比细胞增殖曲线没有明显的变化。结论 :TIMP 2基因转染可以使肿瘤细胞的表面电荷发生变化 ,使细胞的电泳能力、细胞的移动性和侵袭穿透能力减弱 ,而与细胞的生长周期没有明显影响 ,提示TIMP 2基因可能是通过影响细胞的外环境来抑制瘤细胞的恶性生物学表型
AIM: To investigate the effect of matrix metalloproteinase inhibitor (TIMP 2) on the invasiveness of gastric cancer SGC-7901 cells in vitro. Methods: The human full-length TIMP 2 gene was transfected into PA317 cell line by lipofectamine. The cells were screened by G418 and infected with human SGC 790 1 gastric cancer cell line. The cell proliferation, cell electrophoresis rate, soft agar formation and invasion in vitro Analyze its biological behavior change. Results: The SGC-7901 cells and MFC cells and cell electrophoresis rates transfected with TIMP-2 gene were detected. Soft agar formation and invasion ability were significantly lower than the control group, but compared with the control group no significant changes in cell proliferation curve. CONCLUSION: Transfection of TIMP-2 gene can change the surface charge of tumor cells and decrease the cell electrophoretic capacity, cell migration and invasion penetrating ability, but have no significant effect on the cell growth cycle, suggesting that TIMP-2 gene may be Inhibit the malignant biological phenotype of tumor cells by affecting the external environment of cells
