为研究水曲柳合子胚发育后熟时期的基因组表观调控模式,以7月上旬到9月中旬8个取材时期多年生水曲柳合子胚子叶为研究对象,通过基因组甲基化敏感扩增多态性分析,检测了各时期基因组甲基化变化。结果表明:水曲柳合子胚在不同发育时期可检测到933~939条清晰的电泳条带,说明不同发育时期水曲柳合子胚甲基化差异不明显。进一步统计胞嘧啶甲基化模式及其变异模式发现,7月初基因组在CNG、CG/CNG位点下降的变异位点多于其他时期,表明在水曲柳合子胚发育过程中发生了较大规模的去甲基化事件。
