目的 :观察微量元素锌对新生大鼠海马神经元细胞凋亡抑制基因 bcl- 2 m RNA表达的影响 ,探讨锌影响中枢神经系统发育的分子机制。方法 :采用无血清培养 Wistar新生大鼠海马神经元细胞 ,分别在无血清培养液中加入不同浓度的 Zn SO4溶液培养 7d,采用图像分析技术分析其生长分化情况 ;收集海马神经元细胞 ,利用 Northern杂交技术检测海马神经元 bcl- 2m RNA表达情况。结果 :锌在 0 .5~ 4mg/ L 浓度时 ,海马神经元胞体面积、长径、短径以及 bcl- 2 m RNA表达水平均明显高于对照组。 结论 :锌可通过提高 bcl- 2 m RNA转录水平抑制海马神经元细胞凋亡、促进神经元生长发育。
OBJECTIVE: To observe the effect of trace element zinc (Zn) on the expression of bcl-2 mRNA in hippocampal neurons of neonatal rats, and explore the molecular mechanism of zinc affecting CNS development. Methods: The hippocampal neurons of Wistar neonatal rats were cultured in serum-free medium and cultured in serum-free medium with different concentration of ZnSO4 for 7 days respectively. The growth and differentiation of hippocampal neurons were analyzed by image analysis. Detection of bcl-2m RNA expression in hippocampal neurons by hybridization. Results: When the concentration of zinc was 0.5 ~ 4mg / L, the area, long diameter, short diameter and the expression of bcl-2 mRNA in hippocampal neurons were significantly higher than those in the control group. Conclusion: Zinc can inhibit the apoptosis of hippocampal neurons by increasing the transcription level of bcl-2 mRNA and promote the growth and development of neurons.
