目的:观察清心饮对病毒性心肌炎模型的保护作用。方法:取新生 SD 大鼠心室肌制备搏动心肌细胞,18h 后接种100TCID_(50)的柯萨奇 B_3病毒(CVB_3)做为实验性病毒性心肌炎模型。结果:清心饮能够剂量依赖性地减少感染心肌细胞乳酸脱氢酶、天冬氨酸氨基转移酶的释放,降低上清液中的病毒滴度,对心肌细胞搏动异常、停止及细胞病变均有保护作用,且通过斑点杂交发现清心饮能够抑制感染心肌细胞中病毒核糖核酸的复制。结论:临床应用清心饮治疗病毒性心肌炎有实验依据。
Objective: To observe the protective effect of Qingxinyin on viral myocarditis model. Methods: Cardiomyocytes were prepared from ventricular myocardium of neonatal SD rats and inoculated with 100 TCID_(50) of Coxsackie B_3 virus (CVB_3) as an experimental viral myocarditis model after 18 h. RESULTS: Qingxinyin could dose-dependently reduce the release of lactate dehydrogenase and aspartate aminotransferase from infected cardiomyocytes and reduce the titer of virus in supernatant. It has abnormal beat, stoppage and cytopathic effect on myocardial cells. Protective effect, and found by spot hybridization Qing Xin Yin can inhibit viral RNA replication in infected cardiomyocytes. Conclusion: Clinical application of Qingxinyin has experimental basis for the treatment of viral myocarditis.
