目的 :研究半乳糖诱导白内障大鼠异常晶状体蛋白X36的出现及其与白内障形成的关系。方法:半乳糖饲养SD大鼠,诱导白内障;选不同年龄大鼠(出生后1天、2周、8周、8个月及工.5年)作老化进程对照组;提取水溶性晶状体蛋白,双向电泳(等电聚焦/SDS-聚丙烯酞胺凝胶电泳,IEF/SDS-PAGE)得到大鼠晶状体蛋白电泳图形,考马斯亮蓝染色后扫描蛋白图形并确认各主要晶状体蛋白组分及异常晶状体蛋白 X36结果:(1)异常晶状体蛋白X36相对分子质量约 27 000、等电点约4.5。(2)X36蛋白在对照组不同年龄大鼠的水溶性晶状体蛋白中均未出现。(3)X36蛋白在亚期及以后各期糖性白内障大鼠的眼晶状体水溶性蛋白中均有出现,且含量逐期增加。结论:半乳糖诱导大鼠白内障形成过程中出现异常晶状体蛋白X36该蛋白含量随白内障程度加重而增加,与大鼠生理性老化无关。
Objective: To study the appearance of abnormal lens protein X36 in galactose-induced cataract and its relationship with cataract formation. Methods: SD rats were induced by galactose to induce cataract. Rats of different ages (1 day, 2 weeks, 8 weeks, 8 months and 5 years after birth) were selected as control group. Water-soluble lens protein, Two-dimensional electrophoresis (isoelectric focusing / SDS-polyacrylamide gel electrophoresis, IEF / SDS-PAGE) obtained rat lens protein electrophoresis patterns, Coomassie brilliant blue staining protein patterns and confirm the main lens protein components and abnormal lens Protein X36 results: (1) Abnormal lens protein X36 relative molecular mass of about 27 000, the isoelectric point of about 4.5. (2) X36 protein did not appear in water-soluble lens protein of rats of different ages in the control group. (3) The X36 protein appeared in the lens-like water-soluble protein of the cataract rats in the subphase and later stages, and the content increased gradually. CONCLUSION: Galactose induced abnormal lens protein X36 during cataract formation in rats. The protein content increased with the severity of cataract, but not with physiological aging.
