目的:考察灯台叶总生物碱提取物中鸭脚树叶碱和瓦莱萨明碱在大鼠肠道的吸收情况,为其合理用药提供生物药剂学依据。方法:采用大鼠在体肠灌流模型,应用RP-HPLC法测定鸭脚树叶碱及瓦莱萨明碱的含量,研究不同浓度总生物碱中鸭脚树叶碱及瓦莱萨明碱在大鼠不同肠段的吸收情况。结果:相同浓度下鸭脚树叶碱及瓦莱萨明碱在各肠段的有效渗透系数(Peff*)均>1.0,依次为十二指肠>结肠>空肠>回肠,各肠段之间有显著性差异(P<0.05);不同浓度同一肠段,鸭脚树叶碱及瓦莱萨明碱在各肠段的Peff*值无显著性差异。结论:鸭脚树叶碱及瓦莱萨明碱在大鼠肠道四个肠段内均有吸收,尤其在十二指肠、结肠段吸收较好;药物浓度对鸭脚树叶碱及瓦莱萨明碱的Peff*值无显著性影响,初步判断其吸收机制为被动扩散。
OBJECTIVE: To investigate the absorption of duck foot leaf alkali and Valerianamine in the intestinal tract of total alkaloid extract of Xanthoceras sorbifolia leaves, and to provide a biopharmaceutical basis for rational drug use. Methods: Rat intestinal perfused rat model was established. RP-HPLC was used to determine the content of duck foot leaf alkali and Valerianamine. The effects of duck foot leaf alkali and Valerianamine in different concentrations of total alkaloids were studied in rats Absorption of different segments of the bowel. Results: The effective permeability coefficient (Peff *) of duck foot leaf alkali and Valerian spermine in each intestine was> 1.0 at the same concentration, followed by duodenum> colon> jejunum> ileum, and between the intestinal segments (P <0.05). There was no significant difference in the Peff * value of ducks foot leaf alkali and Valerian spermine in the different intestine segments in different intestine segments. CONCLUSION: Both duckweed leaf base and Valerian spermine are absorbed in the intestine of four intestine in rats, especially in the duodenum and colon sections. Peff * value of alkali no significant effect, initially determine the absorption mechanism for passive diffusion.
