用旋光法测定葡萄糖注射液含量时的温度,中国药典85年版规定为20℃。在大于或小于20℃时测得值与在20℃的测得值比较。有何差别,如何处理,目前药典均未提及。为解决这些问题,我们进行如下实验:将10%葡萄糖溶液在不同温度测定其旋光度,并进行回归处理,结果满意。修正值与文献(陈晓龙中国医院药学杂志1983;3(5):234)报道的温度升高1℃,旋光度约减少0.3%不符。现将实验报告如下: 一、材料、仪器原料:注射用葡萄糖(华北制药厂)质量符合药典标准。仪器WZZ自动指示旋光仪(上海光学仪器修理厂)。二、方法与结果精密称取葡萄糖,准确配成10.00%的浓度,分装于100ml的清洁、干燥的盐水瓶中,115℃30分钟灭菌备用。将检品水浴至一定温度后立即测定,用同温度的蒸馏水做空白校正,结果见附表。回归方程y=9.752-0.007597X r=-0.9905P<0.01将实验值按修正值校正后X=100.1%标准差S=0.08721变异系数CV=0.08712%。
