目的:观察赤苷脉通注射液对大鼠急性心肌缺血再灌注损伤的保护作用。方法:采用SD大鼠冠状动脉结扎造成心肌缺血再灌注模型,硝基四氮唑蓝染色法测定大鼠心肌梗死面积,试剂盒测定血清乳酸脱氢酶(LDH)、肌酸激酶(CK)活性,免疫组织化学方法检测大鼠心肌组织中TNF-α及NF-κB p65蛋白表达。结果:赤苷脉通注射液(10,5,2.5 mg/kg)可减小缺血再灌注损伤大鼠心肌梗死面积,抑制大鼠心肌中LDH及CK的漏出,降低血清中LDH与CK的活性,下调缺血再灌注损伤大鼠心肌组织中TNF-α及NF-κB p65蛋白表达。结论:赤苷脉通注射液对大鼠急性心肌缺血再灌注损伤有保护作用,其机制可能与其抗氧化及抑制缺血后炎症反应有关。
OBJECTIVE: To observe the protective effect of ganxin maitong injection on acute myocardial ischemia-reperfusion injury in rats. METHODS: Myocardial ischemia-reperfusion model was established by coronary artery ligation in SD rats. Myocardial infarct size was measured by nitroblue tetrazolium staining. Serum lactate dehydrogenase (LDH) and creatine kinase (CK) were measured by kit. Activity, immunohistochemical method was used to detect the expression of TNF-α and NF-κB p65 protein in rat myocardial tissue. RESULTS: The injection of Lanshen Maitong (10,5,2.5 mg/kg) reduced the myocardial infarct size in rats with ischemia-reperfusion injury, inhibited the leakage of LDH and CK in the myocardium of rats, and decreased the levels of LDH and CK in serum. Activity, down-regulation of TNF-α and NF-κB p65 protein expression in myocardial tissue of rats with ischemia-reperfusion injury. Conclusion: The injection of Lanshen Maitong has a protective effect on acute myocardial ischemia-reperfusion injury in rats, and its mechanism may be related to its anti-oxidation and inhibition of inflammation after ischemia.
