目的 研究维拉帕米( 异博定,钙离子阻滞剂)、双嘧达莫( 潘生丁) 、丹参、黄体酮、环孢菌素A、他莫昔芬等药物对人卵巢癌细胞系( 具有多药耐药的特征) 顺铂(DDP) 耐药性的逆转作用。 方法 用MTT 法测定上述药物对A2780/DDP耐药细胞的细胞毒性作用,用高效液相色谱仪(HPLC) 测定细胞内DDP 含量。 结果 除丹参以外的其他5 种药物,能不同程度地逆转耐药细胞对顺铂的耐药性;可增加耐药细胞内顺铂的药物浓度。 结论 耐药细胞逆转作用主要是由于这些药物使耐药细胞内对顺铂解毒系统功能降低,致使细胞内药物浓度增加,与多药耐药基因( MDR1) 关系不大。
Objective To study the effects of verapamil (isoboridine, calcium blockers), dipyridamole (dipyridamole), salvia miltiorrhiza, progesterone, cyclosporine A and tamoxifen on human ovarian cancer cell line ( Multidrug resistance characteristics) DDP reversal of drug resistance. Methods MTT assay was used to determine the cytotoxicity of the above drugs on A2780 / DDP-resistant cells. The content of intracellular DDP was determined by high performance liquid chromatography (HPLC). Results In addition to Danshen other than 5 kinds of drugs, to varying degrees, reversal of drug resistance of cisplatin-resistant cells; drug-resistant cells can increase the concentration of cisplatin. Conclusion The reversal effects of drug-resistant cells are mainly attributed to the decreased function of cisplatin detoxification system caused by these drugs in drug-resistant cells, resulting in the increase of intracellular drug concentration, which has little to do with multidrug resistance gene (MDR1).
