目的 比较 3种汽油增氧剂对小鼠成纤维细胞DNA的损伤作用。方法 采用单细胞凝胶电泳试验 (彗星试验 )对 3种汽油增氧剂甲基叔丁基醚 (MTBE)、无水乙醇 (EA)和碳酸二甲酯 (DMC)所致的L 92 9小鼠成纤维细胞的DNA损伤进行了研究。结果 在一定浓度范围内 (37.5 0 0~ 15 0 .0 0 0mg/ml) ,MTBE可直接引起L 92 9小鼠成纤维细胞的DNA损伤 ,出现拖尾的彗星细胞。其拖尾细胞百分率和DNA迁移长度均随受试物浓度的增加而增加 ,且有明显的剂量 -效应关系 ,即MTBE浓度从9.375mg/ml增加到 15 0 .0 0 0mg/ml时 ,彗星率从接近阴性对照组的 4 %增加到接近阳性对照组的85 % ,彗尾长度也发生了相应的改变。而EA和DMC无此关系。结论 在本试验条件下 (浓度为15 0 .0 0 0mg/ml) ,MTBE对DNA具有明显的损伤作用 ;未发现EA和DMC对DNA的损伤作用。
Objective To compare the DNA damage effects of three kinds of gasoline aerobes on mouse fibroblasts. Methods Single cell gel electrophoresis (Comet assay) was used to investigate the effects of three petrol oxygenators methyl tert-butyl ether (MTBE), absolute ethyl alcohol (EA) and dimethyl carbonate (DMC) DNA damage in murine fibroblasts was studied. Results MTBE could directly induce the DNA damage of L 92 9 mouse fibroblasts in a certain concentration range (37.5 0 0 ~ 150.000 mg / ml), and the trailing comet cells appeared. The percentage of tailing cells and the length of DNA migration increased with the increase of the concentration of the test substance, and there was a significant dose-effect relationship, that is, when the concentration of MTBE increased from 9.375mg / ml to 150.0mg / ml, comet The rate increased from 4% near the negative control group to nearly 85% of the positive control group, and the tail length also changed correspondingly. EA and DMC have no such relationship. Conclusion MTBE has a significant DNA damage effect under the experimental conditions (concentration of 150.0 mg / ml). No DNA damage was observed by EA or DMC.
