目的:观察丹皮不同提取物对子宫内膜异位症(endometriosis,EMS)模型动物血液流变学、血清IL-1、IL-6含量以及病理组织的影响,以期探讨丹皮治疗的EMS的物质基础及部分作用机制。方法:采用手术移植法制备大鼠子宫内膜异位症模型,并随机分成模型组、桂枝茯苓胶囊组、丹皮水提物组、丹皮酚组、丹皮多糖组、丹皮总苷组,并设假手术组。连续给药30 d后,麻醉动物,腹主动脉采血,测定血液流变学指标;分离血清,采用双抗体夹心ABC-ELISA法测定血清IL-1、IL-6含量。摘取异位子宫内膜组织,10%中性福尔马林固定,HE染色进行病理学检查。结果:模型动物的全血粘度、血浆粘度以及血清中IL-1、IL-6含量均较假手术组显著增加(P<0.05);不同丹皮提取物均能改善模型动物异位子宫内膜的病理组织学改变,抑制基质细胞的增生;对血液粘度均有一定的改善作用,但与模型组比较无显著性差异(P>0.05)。丹皮酚、丹皮总苷能显著降低模型动物血清IL-1、IL-6含量(P<0.05)。结论:丹皮不同提取物对EMS有一定的治疗作用,其机制可能与降低IL-1、IL-6含量,减轻炎症反应有关,其物质基础可能为丹皮酚及丹皮总苷。
OBJECTIVE: To observe the effects of different extracts of Radix Paeoniae Alba on blood rheology, serum IL-1 and IL-6 levels and pathological tissues in endometriosis (EMT) model animals in order to investigate the effects of paeonol treatment on EMS Material basis and part of the mechanism. Methods: The rat model of endometriosis was prepared by surgical transplantation and randomly divided into three groups: model group, Guizhi Fuling capsule group, water extract of Dandelion group, paeonol group, Dan Pill polysaccharide group, Group, and set the sham operation group. After continuous administration for 30 days, blood was collected from the anesthetized animals and the abdominal aorta to determine the indexes of hemorrheology. Serum levels of IL-1 and IL-6 were measured by double antibody sandwich ABC-ELISA. Ectopic endometrial tissues were removed, fixed in 10% neutral formalin, and stained with HE for pathological examination. Results: The whole blood viscosity, plasma viscosity and serum IL-1 and IL-6 levels in model animals were significantly increased compared with those in sham operation group (P <0.05). Different extracts of Bupleurum improved the ectopic endometrium of model animals The histopathological changes and the proliferation of stromal cells inhibited the blood viscosity, but there was no significant difference compared with the model group (P> 0.05). Paeonol and tamponin could significantly reduce the levels of IL-1 and IL-6 in model animals (P <0.05). Conclusion: Different extracts of Radix Paeoniae Rubra have a certain therapeutic effect on EMS. The mechanism may be related to the decrease of IL-1 and IL-6 content and the reduction of inflammatory reaction. The substance basis may be paeonol and total anthocyanin.
