目的:研究款冬花、紫菀配伍对绿原酸在大鼠体内药代动力学参数的影响。方法:在筛选款冬花止咳有效部位、紫菀止咳增效部位的基础上,将SD大鼠随机分成4组,分别灌胃给予绿原酸单体、款冬花部位、紫菀部位、款冬花部位+紫菀部位(即部位配伍),采用HPLC法测定血浆中的绿原酸浓度,并用WinNonlin药动学软件根据非房室模型法计算药动学参数。结果:大鼠灌胃绿原酸单体、款冬花部位、紫菀部位、部位配伍后的主要药动参数:AUC0-t分别为245.09±14.05,349.27±62.15,105.11±18.57,496.77±77.75μg.mL-1.min-1;t1/2分别为323.70±51.00,464.53±126.10,408.27±31.30,203.02±7.12min;Cmax分别为1.05±0.13,1.52±0.55,0.63±0.04,4.49±0.59μg.mL-1;Tmax分别为60.00±0.00,15.00±0.00,15.00±0.00,15.00±0.00min。结论:紫菀能促进大鼠对款冬花中绿原酸的吸收,加快绿原酸的消除,减少绿原酸在体内的蓄积,可以部分诠释款冬花、紫菀“相须配伍”增效的机理。
OBJECTIVE: To study the effects of Coltsfoot flower and Aster on the pharmacokinetic parameters of chlorogenic acid in rats. Methods: On the basis of selecting effective part of antitussive cough and antitussive and cough synergistic parts, SD rats were randomly divided into 4 groups, and were given gavage of chlorogenic acid monomer, anther colchicum part, aster columbine part + aster (Ie site compatibility). The concentration of chlorogenic acid in plasma was determined by HPLC. Pharmacokinetic parameters were calculated by non-compartmental model with WinNonlin pharmacokinetic software. Results: The main pharmacokinetic parameters of rats after gavage of chlorogenic acid monomer, coltsfoot flower and purple aspartame were: AUC0-t of 245.09 ± 14.05,349.27 ± 62.15,105.11 ± 18.57,496.77 ± 77.75μg. mL-1.min-1; t1 / 2 was 323.70 ± 51.00, 464.53 ± 126.10, 408.27 ± 31.30 and 203.02 ± 7.12min, respectively; Cmax was 1.05 ± 0.13, 1.52 ± 0.55, 0.63 ± 0.04 and 4.49 ± 0.59μg, respectively. mL-1; Tmax was 60.00 ± 0.00,15.00 ± 0.00,15.00 ± 0.00,15.00 ± 0.00min, respectively. CONCLUSION: Aster can promote the absorption of chlorogenic acid in Coltsfoot flower, accelerate the elimination of chlorogenic acid, and reduce the accumulation of chlorogenic acid in the body, which can partly explain the synergistic effects of coltsfoot flower, aster syrup mechanism.
