本实验用培养的家兔血管平滑肌细胞探讨了在促进细胞增生过程中,氧化低密度脂蛋白(oxidizedlowdensitylipoprotein,OLDL)和内皮素之间的联系,发现二者都可刺激血管平滑肌细胞增生,内皮素A受体拮抗剂BQ123可显著抑印制OLDL的促细胞增生作用, ̄3H-TdR的掺入量较单纯OLDL组减少13.3%;细胞培养液内皮素放射免疫测定表明,应用BQ123能使内皮素释放量较单纯OLDL组降低28.9%。免疫细胞化学检测结果亦与上述完全符合,提示在动脉粥样硬化发生中,OLDL促血管平滑肌细胞增生的作用可能部分是通过内皮素来实现的。
In this study, rabbit vascular smooth muscle cells were cultured to explore the promotion of cell proliferation, oxidized low density lipoprotein (oxidized low density lipoprotein (OLDL) and the relationship between endothelin and found that both can stimulate vascular smooth muscle cell proliferation, endothelin A receptor antagonist BQ123 significantly inhibited the proliferation of cells in the role of OLDL, ~ 3H-TdR incorporation than the simple OLDL group decreased by 13.3%; cell culture fluid endothelin radioimmunoassay showed that the application of BQ123 can make Endothelin release than the simple OLDL group decreased by 28.9%. The results of immunocytochemistry were in good agreement with the above findings, suggesting that the effect of OLDL on the proliferation of vascular smooth muscle cells may be partially mediated by endothelin in the development of atherosclerosis.
