目的探索一测多评法同时测定桔梗中3种桔梗皂苷含量的可行性。方法采用HPLC-ELSD为检测手段,考察桔梗提取物在不同色谱系统和不同色谱柱上的色谱行为,确定3种待测成分的色谱峰定位方法。以桔梗皂苷D为内标,建立其与桔梗皂苷D_3、桔梗皂苷E的相对校正因子;通过外标法测定桔梗皂苷D的含量,并由建立的相对校正因子计算出其他2种桔梗皂苷的含量;将计算的结果与外标法测定结果相比较,以验证“一测多评”法在桔梗质量评价中的可行性。结果方法重复性和耐用性良好,在10批桔梗药材的含量测定中,一测多评法计算结果与外标法测定结果无显著差异。结论一测多评法可用于同时测定桔梗中多种皂苷的含量。
Objective To explore the feasibility of simultaneous determination of three kinds of platycodin in Platycodon grandiflorum by multi-evaluation method. Methods HPLC-ELSD was used as the detection method to investigate the chromatographic behavior of Campanulaceae extracts on different chromatographic systems and different chromatographic columns, and to determine the chromatographic peak locating methods of the three tested components. Platycodin D was used as internal standard to establish its relative correction factor with platycodin D_3 and Platycodin E; the content of Platycodin D was determined by external standard method, and the content of other platycodin was calculated from the established relative correction factor The calculated results were compared with the results of external standard method to verify the feasibility of using the method of multiple test and multiple test in the quality evaluation of Campanulaceae. Results The repeatability and durability of the method were good. In the determination of the content of 10 samples of Platycodon, there was no significant difference between the results of one method and those of external standard method. Conclusion A multi-evaluation method can be used to simultaneously determine a variety of Campanulaceae saponin content.
