目的研究大孔吸附树脂富集二黄胶囊水提部分中5-羟甲基糠醛的工艺条件及参数。方法以5-羟甲基糠醛为指标,通过静态及动态吸附指标的考察,确定了筛选的树脂型号;采用单因素实验考察了吸附时间、上样量与树脂体积比;采用正交实验,对工艺参数进行了优化。结果正交实验表明,影响二黄胶囊大孔树脂精制工艺的主要因素为树脂柱径高比和药液浓度,其最佳工艺条件为生药量与树脂体积比为1∶4,柱床径高比为1∶8,上样药液浓度为1∶8,吸附流速为1 mL/min。结论5-羟甲基糠醛过树脂前后含量保留90%以上,用大孔树脂对二黄胶囊水提液进行精制工艺是合理可行的。
Objective To study the process conditions and parameters for enrichment of 5-hydroxymethylfurfural in the aqueous extract of Erhuang Capsule by macroporous adsorption resin. Methods 5-Hydroxymethylfurfural was used as an index to determine the resin type by static and dynamic adsorption. The adsorption time, the amount of sample and the resin volume ratio were investigated by single factor experiment. By orthogonal experiment, Process parameters are optimized. Results Orthogonal experiments showed that the main factors influencing the purification process of Erhuang capsule macroporous resin were resin column diameter-height ratio and liquid concentration. The optimal process conditions were the ratio of crude drug to resin volume ratio of 1: 4, Ratio of 1: 8, sample solution concentration of 1: 8, adsorption flow rate of 1 mL / min. Conclusion The content of 5-hydroxymethylfurfural in the resin is more than 90%. It is reasonable and feasible to use the macroporous resin to purify the water extract of Erhuang Capsule.
