目的:通过检测大鼠肾功能指标和血清醛固酮、钠钾离子的含量,初步观察醋甘遂与炙甘草免煎颗粒配伍对肾脏的损伤。方法:将大鼠按体重随机分为正常组、炙甘草组(免煎颗粒剂)、醋甘遂组(研末)、醋甘遂-炙甘草组、甘遂半夏汤组。其中正常组给予生理盐水灌胃,其余各组给予相应药液灌胃,连续给药14天。实验结束时,腹主动脉取血分离血清和血浆检测肌酐(Cr)、尿素氮(BUN)、尿酸(UA)、乳酸脱氢酶(LDH)、醛固酮(ALD)的含量以及血清中Na+、K+含量;取一侧肾组织用福尔马林固定观察肾组织细胞病理形态。结果:与正常组相比,炙甘草组、醋甘遂组、醋甘遂-炙甘草组Cr含量有升高趋势,但差异无统计学意义(P>0.05),BUN含量各组均有升高趋势,醋甘遂-炙甘草组升高显著,差异有显著统计学意义(P<0.01),且与醋甘遂组比较,亦显著升高,差异有统计学意义(P<0.05)。尿酸含量各组之间无明显变化,LDH含量显著降低,差异有统计学意义(P<0.05,P<0.01)。血清Na+、K+含量:与正常组比较,各给药组之间无显著性变化,差异无统计学意义(P>0.05)。醛固酮(ALD)含量:与正常组比较,各给药组均显著降低,差异有统计学意义(P<0.05,P<0.01)。其中甘遂半夏汤组较醋甘遂-炙甘草组显著升高,差异有统计学意义(P<0.05)。结论:甘遂确实对肾脏造成了损伤,但醋甘遂与炙甘草配伍未表现出明显的毒性增强作用,说明二者配伍不存在增毒作用,是否有增效作用需要配合病理状态的研究来深入探讨。
OBJECTIVE: To observe the kidney function of rats and the contents of aldosterone, sodium and potassium in serum and to observe the renal damage caused by the compatibility of Zhuguishao and Zhigancao decoction. Methods: The rats were randomly divided into normal group, Zhigancao group (decocted granules), vinegar group (end of the powder), vinegar Gan Sui - Zhigancao group, Gan Sui Banxia Tang group. The normal group was given normal saline gavage, and the remaining groups were given the corresponding liquid gavage, continuous administration for 14 days. Serum creatinine (Cr), blood urea nitrogen (BUN), uric acid (UA), lactate dehydrogenase (LDH) and aldosterone (ALD) were detected in serum and plasma of abdominal aorta at the end of the experiment. Content; take a side of kidney tissue with formalin fixed observation renal cell pathology. Results: Compared with the normal group, the contents of Cr in Zhigancao, Zhugansuai and Zhugansui - Zhigancao groups had an increasing tendency, but the difference was not statistically significant (P> 0.05). The content of BUN increased in all groups (P <0.01). Compared with those in the vinegared group, the levels were also significantly increased (P <0.05) .Conclusion1. Uric acid content between the groups showed no significant change, LDH content was significantly lower, the difference was statistically significant (P <0.05, P <0.01). Serum Na +, K + content: compared with the normal group, there was no significant difference between each administration group, the difference was not statistically significant (P> 0.05). Aldosterone (ALD) content: compared with the normal group, each treatment group were significantly lower, the difference was statistically significant (P <0.05, P <0.01). Among them, the Kansui Banxia Decoction group was significantly higher than that of the vinegar Kampuchi - Zhigancao group, the difference was statistically significant (P <0.05). Conclusion: Kansui really caused damage to the kidneys. However, the combination of vinegar and kundsu with Zhigancao did not show significant toxicity enhancement, indicating that there is no toxic effect between the two compatibility, and whether there is synergism need to cooperate with the pathological state discuss in depth.
