目的 :观察尾加压素Ⅱ (UⅡ )对心肌细胞 (MC)肥大的影响及其与钙离子的关系 ,为探讨高血压左室肥厚发生机制提供理论依据 .方法 :以培养的SD乳鼠心肌细胞为实验模型 ,通过测定MC表面积、蛋白含量和蛋白合成速率 ,以及免疫荧光观察MC内肌原纤维的变化趋势来探讨UⅡ对MC肥大的影响 .结果 :①随着UⅡ浓度的增加 ,MC表面积明显增加 ,呈剂量依赖性 ,其中 1 0 -8和 1 0 -7mol/LUⅡ组MC表面积分别为 (2 0 4 6 .3± 2 6 8.4 )和 (36 6 2 .2± 2 5 9.2 ) μm2 ,均明显高于空白对照组的 (936 .2± 99.8) μm2 ,差异有非常显著意义 (P <0 .0 1 ) .②随着UⅡ浓度的增加 ,MC的蛋白含量和蛋白合成速率呈递增趋势 ,其中 1 0 -8和 1 0 -7mol/LUⅡ组MC蛋白含量分别为 (1 .6 0± 0 .37)和 (1 .89± 0 .2 5 )ng/cell,明显高于对照组(0 .83± 0 .1 6 )ng/cell,差异有非常显著性意义 (P <0 .0 1 ) .蛋白合成速率分别为 (2 5 0 8.33± 2 99.81 )和 (2 898.83± 6 2 5 .4 1 )cpm/well,与对照组 (1 0 2 6 .83± 91 .6 7)cpm/well比较 ,差异非常显著 (P <0 .0 1 ) .③ 1 0 -7mol/LUⅡ作用心肌细胞 2 4h后 ,荧光显微镜下观察到细胞边界增大 ,肌原纤维发生增粗与重排 .结论 :UⅡ能够诱导心肌细胞的肥大 ,为研究高血压及其他心血管?
Objective: To investigate the effect of urotensin Ⅱ (UⅡ) on the hypertrophy of cardiomyocytes (MC) and its relationship with calcium ion in order to provide a theoretical basis for exploring the mechanism of hypertensive left ventricular hypertrophy.Methods: Cells as experimental model to explore the effect of UⅡ on MC hypertrophy by measuring the surface area, protein content and protein synthesis rate of MC, as well as the changes of myofibrils in immunofluorescence.Results: ①With the increase of UⅡ concentration, the MC surface area (2 0 4 6 3 ± 2 6 8.4) and (36 6 2 2 2 5 9.2) μm 2 in the groups of 1 0 -8 and 1 0 -7mol / LUⅡ, respectively , Which were significantly higher than that of the blank control group (936.2 ± 99.8) μm2, the difference was significant (P0.01) .②With the increase of UⅡ concentration, the protein content and protein synthesis rate of MC increased gradually (1.06 ± 0.37) and (1.89 ± 0.25) ng / cell respectively, which was significantly higher than that of the control group (0.83 ± 0.16) ng / cell, the difference was significant (P <0.01) .The rate of protein synthesis was (2 58.33 ± 2 99.81) and (2 898.83 ± 6 2 5 4 1) cpm / well, compared with the control group (1 0 2 6 .83 ± 91 6 7 cpm / well), the difference was significant (P 0 01) .③ 1 After treated with 0 ~ 7 mol / L of cardiomyocytes for 24 hours, the cell borders were enlarged and myofibrils were thicker and rearranged under the fluorescence microscope.Conclusion: UⅡ can induce the hypertrophy of cardiomyocytes, ?
