目的 观察乙型肝炎病毒核心抗原核酸疫苗即 HBc Ag核酸疫苗 (p JW430 3/ HBc)免疫猕猴的特异性细胞免疫应答。方法 猕猴分别在第 0、2、4、6个月肌肉注射法接种核酸疫苗 (p JW430 3/ HBc)或对照质粒 (p JW430 3) ;EL ISA法检测猕猴外周血单个核细胞 (PBMC) ,经特异性抗原 (HBc Ag)刺激后 ,培养上清中人白细胞介素 - 4(IL- 4)和干扰素 γ(IFN- γ)以及猕猴血清中抗 - HBc Ig G亚类水平 ;3H- TDR掺入法测定猕猴 PBMC特异性增殖反应。结果 接种核酸疫苗后猕猴 PBMC培养上清中以IFN- γ升高为主。血清中特异性抗体 (抗 - ΗΒc) Ig G亚类以 Ig G2占优势。免疫组猕猴 PBMC抗原特异性增殖反应明显增强 ,增殖指数达 3.83,与对照猕猴相比有显著差异。结论 p JW430 3/ HBc核酸疫苗能有效诱导猕猴的特异性细胞免疫应答
Objective To observe the specific cellular immune responses of monkeys immunized with hepatitis B virus core antigen nucleic acid vaccine, HBcAg nucleic acid vaccine (p JW430 3 / HBc). Methods The rhesus monkeys were inoculated intramuscularly with pJW430 3 / HBc or pJW430 3 at the 0, 2, 4, 6 months respectively. The peripheral blood mononuclear cells (PBMC) After stimulation with specific antigens (HBc Ag), the levels of human interleukin-4 (IL-4) and interferon gamma (IFN-γ) and the anti-HBc Ig G TCR incorporation method was used to determine the PBMC specific proliferative response in rhesus monkeys. Results After inoculation of nucleic acid vaccine, the increase of IFN-γ in the supernatant of PBMC of rhesus monkeys was predominant. Serum specific antibodies (anti-ΗΒc) Ig G subtype dominated by Ig G2. Immunization group PBMC antigen-specific proliferative response was significantly enhanced, the proliferation index of 3.83, compared with the control rhesus monkey significant difference. Conclusion p JW430 3 / HBc nucleic acid vaccine can effectively induce the specific cellular immune response
