以10年生马占相思和大叶相思优良单株为研究对象,其茎段外植体通过初代培养和增殖培养后获得增殖芽,研究了生长素种类及浓度、基本培养基类型、蔗糖浓度和活性炭浓度对增殖芽生根的影响,分析了不同类型增殖培养基多次继代诱导的增殖芽对生根的影响。结果表明:马占相思的最佳生根培养基为1/2MS+0.5 mg/L IBA+0.5 mg/L NAA+3%蔗糖,大叶相思最佳生根培养基为1/2MS+0.5 mg/L IBA+0.25 mg/L NAA+3%蔗糖,二者的生根率均能达到93%以上。以马占相思和大叶相思增殖芽为生根材料,含低浓度细胞分裂素且添加活性炭的增殖培养基诱导时,两种相思的生根率均能达到90%以上,生根率随着继代次数的增加保持稳定,生根苗生长健壮。
