目的探讨海昆肾喜对糖尿病肾病大鼠肾组织转化生长因子-β1(TGF-β1)蛋白及mRNA表达的影响。方法50只大鼠空腹一次性腹腔注射链脲佐菌素造成糖尿病模型,并按血糖高低随机分为2组、试验组灌胃海昆肾喜,给药12周。第4、8、12周末收集大鼠24h尿量及检测尿蛋白量。12周末处死大鼠,取血并分离血清检测肌酐(Scr)、尿素氮(BUN)和甘油三酯(TG)等,取肾后固定包埋切片,采用HE、Mallory染色观察肾组织病理改变,并应用免疫组化检测肾组织TGF-β1及用原位杂交检测肾组织TGF-β1mRNA的表达。结果海昆肾喜能够降低糖尿病大鼠的蛋白尿,增加体重,降低血清Scr和BUN,改善肾功能,调节TG,改善脂代谢紊乱,并且能够降低糖尿病大鼠肾小管上皮细胞、肾间质细胞浆TGF-β1及mRNA的表达。结论海昆肾喜可能抑制TGF-β1蛋白及mRNA的过度表达,进而减缓糖尿病肾病的病程进展。
Objective To investigate the effect of Haikun Shenxi on the protein and mRNA expression of transforming growth factor-β1 (TGF-β1) in the kidney of diabetic nephropathy rats. Methods Fifty rats were injected intraperitoneally with streptozotocin (STZ) once a day to induce diabetic rats. The rats were randomly divided into 2 groups according to their blood glucose levels. At the end of the 4th, 8th and 12th week, the urinary excretion of urine and the amount of urine protein were collected. The rats were sacrificed on the 12th week, blood was drawn and the serum was collected for determination of creatinine (Scr), blood urea nitrogen (BUN) and triglyceride (TG), fixed and embedded in the kidney. HE and Mallory staining were used to observe the pathological changes, Immunohistochemistry was used to detect TGF-β1 in renal tissue and the expression of TGF-β1 mRNA in renal tissue was detected by in situ hybridization. Results Hai Kun Shen Xi can reduce diabetic rats proteinuria, increase body weight, lower serum Scr and BUN, improve renal function, regulate TG, improve lipid metabolism disorder, and can reduce the diabetic rat renal tubular epithelial cells, renal interstitial cells Plasma TGF-β1 and mRNA expression. Conclusion Haikun Shenxi may inhibit TGF-β1 protein and mRNA overexpression, thereby reducing the course of diabetic nephropathy.
