目的:通过观察大鼠局灶性脑缺血再灌注后脑功能行为及脑组织形态学改变,研究川芎嗪和葛根素配伍对脑缺血再灌注损伤的保护作用机制。方法:采用改良的Pulsinelli四血管法建立大鼠脑缺血损伤模型,将SD大鼠随机分为假手术组、模型组、川加葛组(川芎嗪50 mg.kg-1和葛根素50 mg.kg-1)、尼莫地平组(0.5 mg.kg-1),于再灌0,6 h ip。用药后,观察药物对脑缺血引起的神经功能缺损症状、脑组织形态学变化的影响。结果:造模后大鼠大脑皮质脑组织形态学结构发生改变及神经元损害等改变。川芎嗪与葛根素配伍不仅能减轻模型大鼠的神经功能缺损症状,且大鼠大脑皮质神经元病变有一定程度的改善。结论:川芎嗪和葛根素配伍对脑缺血损伤有一定防治作用。
OBJECTIVE: To observe the protective effect of ligustrazine and puerarin on cerebral function and brain morphological changes after focal cerebral ischemia-reperfusion in rats. Methods: A rat model of cerebral ischemic injury was established by a modified Pulsinelli four-vessel method. The SD rats were randomly divided into sham operation group, model group and chuanghgal group (ligustrazine 50 mg.kg-1 and puerarin 50 mg .kg-1), nimodipine group (0.5 mg.kg-1) at 0 and 6 h after reperfusion. After treatment, the effects of drugs on neurological deficits caused by cerebral ischemia and brain morphological changes were observed. Results: After the model was established, the morphological changes of cerebral cortex in rats and neuronal damage were also observed. The combination of ligustrazine and puerarin can not only alleviate the symptoms of neurological deficit in the model rats, but also improve the neuronal lesions of the cerebral cortex in rats to some extent. Conclusion: The combination of ligustrazine and puerarin has some preventive and therapeutic effects on cerebral ischemia.
