目的为研究消化道染镉对雄性大鼠生殖机能毒性作用的机制,本实验观察了饮水染镉对大鼠血浆和睾丸组织中生殖激素的影响。方法 15只雄性SD大鼠(性未成熟)随机分成Ⅰ、Ⅱ和Ⅲ共3组,每组5只。Ⅰ组为对照组,饮用正常清洁饮用水;Ⅱ组、Ⅲ组分别自由饮用添加Cd浓度分别为50和200 mg/L的氯化镉溶液的饮用水,试验期为105 d。后用免疫放射法(RIA法)分别检测了血浆中的睾酮(T)、卵泡刺激素(FSH)、促黄体生成素(LH)和睾丸组织中的睾酮(T)含量。结果随着染镉剂量的增加,大鼠血浆中T的水平呈下降趋势,染镉组与对照组、染镉组间差异均有统计学意义(P<0.01);血浆中FSH和LH的水平呈升高趋势,各组间差异均有统计学意义(P<0.01);睾丸组织中T的水平呈下降趋势,各组间差异也均有统计学意义(P<0.01)。结论饮水染镉条件下,镉对雄性动物睾丸睾酮的合成和分泌具有抑制作用,至于血浆中FSH和LH的升高,可能是反馈调节的结果。
Objective To study the mechanism of toxic effects of cadmium in the digestive tract on reproductive function in male rats. The effects of cadmium in drinking water on reproductive hormones in plasma and testis of rats were observed. Methods Fifteen male SD rats (immature) were randomly divided into three groups Ⅰ, Ⅱ and Ⅲ, five in each group. Group Ⅰ was the control group, drinking normal clean drinking water; Group Ⅱ, Ⅲ were free to drink cadmium chloride solution with Cd concentrations of 50 and 200 mg / L drinking water, the test period was 105 d. The contents of testosterone (T), follicle stimulating hormone (FSH), luteinizing hormone (LH) and testosterone (T) in testis were detected by radioimmunoassay (RIA). Results With the increase of cadmium dose, the level of T in plasma decreased. There was significant difference between cadmium group and control group and cadmium group (P <0.01). The level of plasma FSH and LH (P <0.01). The level of T in testicular tissue showed a decreasing trend and the differences among the groups were also statistically significant (P <0.01). Conclusions Cadmium has an inhibitory effect on the synthesis and secretion of testosterone in testis in male pigs exposed to cadmium in drinking water. As for the increase of FSH and LH in plasma, it may be the result of feedback regulation.
