目的:研究扶正固本方对结肠癌LOVO细胞株在体外增殖的抑制作用及其机制。方法:用不同浓度的扶正固本方水提物分别作用于对数期的LOVO细胞24小时及48小时,通过透射电镜观察细胞形态的变化并应用四甲基偶氮唑蓝(MTT)实验检测细胞的增殖抑制率,流式细胞术检测细胞周期。结果:随药物浓度的增加和作用时间的延长,扶正固本方对lOVO细胞的杀伤作用逐渐增强(P<0.05)。作用48小时后,扶正固本方不同浓度含药组与空白组比较,G0/G1期细胞含量明显增多(P<0.01),S期细胞含量明显减少(P<0.01),以含药高浓度组作用最为明显。结论:扶正固本方水提物可有效抑制结肠癌LOVO细胞的体外增殖,其机制与干扰S期细胞DNA合成有关。
Objective: To study the inhibitory effect of Fuzheng Gubenfang on the proliferation of colon cancer LOVO cell lines in vitro and its mechanism. Methods: The LOVO cells in different logarithmic phases were treated with different concentrations of Fuzhengganzhu water extract for 24 hours and 48 hours respectively. The morphological changes of the cells were observed by transmission electron microscopy and detected by MTT assay Cell proliferation inhibition rate, flow cytometry cell cycle. Results: With the increase of drug concentration and prolongation of action time, the killing effect of Fuzheng Guben Formula on lOVO cells gradually increased (P <0.05). After 48 hours, the content of G0 / G1 phase cells in Fuzheng Guben prescription group and control group were significantly increased (P <0.01), while the content of S phase cells was significantly decreased (P <0.01) The role of the group most obvious. Conclusion: Fuzheng Gubenfang aqueous extract can effectively inhibit the proliferation of colon cancer LOVO cells in vitro, the mechanism of which is related to the interference of DNA synthesis in S phase cells.
