目的:研究胃癌中SLC7A5转录和表达的特点,探讨SLC7A5对胃癌细胞增殖的影响及其在胃癌发生、发展中的临床意义。方法:应用免疫组化法检测52对临床采集的胃癌组织及相应的癌旁组织中SLC7A5的表达情况;分析4株胃癌细胞(SGC-7901、MKN-45、MGC-803和CRL-5974)中SLC7A5转录及表达水平与永生化胃黏膜细胞GES-1间的差异;结合临床病理指标,分析SLC7A5在胃癌组织中的表达水平与各项临床病理特征间的相关性;并应用pGU6/GFP/Neo/shRNA-SLC7A5质粒载体转染胃癌SGC-7901细胞,下调SLC7A5表达,通过CCK8检测及流式细胞术检测分析SLC7A5对胃癌细胞增殖和细胞周期的影响。结果:SLC7A5在胃癌组织和细胞中的表达上调,且其表达水平与胃癌肿瘤的直径、局部浸润深度、淋巴结转移及TNM分期进展显著相关;而下调胃癌SGC-7901细胞SLC7A5表达,可显著减弱胃癌细胞增殖能力,并将细胞周期阻滞于G0/G1期。结论:SLC7A5可促进胃癌细胞增殖,与其肿瘤直径、局部浸润深度、淋巴结转移及TNM分期等临床病理特征相关,可作为分子标志物评价胃癌的生物学行为,并可能成为胃癌诊断和防治中的新靶点。
OBJECTIVE: To study the characteristics of SLC7A5 transcription and expression in gastric cancer and to investigate the effect of SLC7A5 on the proliferation of gastric cancer cells and its clinical significance in the occurrence and development of gastric cancer. Methods: Immunohistochemical method was used to detect the expression of SLC7A5 in 52 gastric cancer tissues and corresponding paracancerous tissues. The expressions of SLC7A5 in 4 gastric cancer cells (SGC-7901, MKN-45, MGC-803 and CRL-5974) SLC7A5 expression and immortalized gastric mucosal cells GES-1 differences; combined with clinical and pathological indicators, the expression of SLC7A5 in gastric cancer and the relationship between the various clinical and pathological features; and the application of pGU6 / GFP / Neo / shRNA-SLC7A5 plasmid was transfected into gastric cancer SGC-7901 cells to down-regulate the expression of SLC7A5. The effects of SLC7A5 on the proliferation and cell cycle of gastric cancer cells were analyzed by CCK8 assay and flow cytometry. Results: The expression of SLC7A5 in gastric cancer tissues and cells was up-regulated, and the expression level of SLC7A5 was significantly correlated with tumor diameter, depth of invasion, lymph node metastasis and TNM staging. However, SLC7A5 expression was significantly decreased in gastric cancer SGC-7901 Cell proliferation ability, and cell cycle arrest in G0 / G1 phase. Conclusion: SLC7A5 can promote the proliferation of gastric cancer cells, which is related to clinicopathological features such as tumor diameter, depth of invasion, lymph node metastasis and TNM staging. It can be used as a molecular marker to evaluate the biological behavior of gastric cancer and may become a new diagnosis and control of gastric cancer Targets.
