目的:研究翠云草总黄酮对结肠癌HT-29细胞环氧化酶-2(COX-2)mRNA表达的影响。方法:以翠云草总黄酮作用于人结肠癌HT-29细胞,用倒置显微镜观察细胞形态学的变化;并设总黄酮高中低剂量组和阴性对照组共4个组别,采用逆转录聚合酶链反应(RT-PCR)法检测给药后癌细胞COX-2 mRNA表达的水平,用电泳凝胶成像自动分析系统检测各扩增带的产物含量。结果:与阴性对照组比较,翠云草总黄酮可以在mRNA水平上抑制COX-2的表达,且呈量效关系。结论:翠云草总黄酮能在mRNA水平上抑制COX-2,进而抑制COX-2蛋白质表达,这可能是翠云草抗肿瘤作用的机制之一。
Objective: To investigate the effect of total flavonoids of Cuiyucao on the expression of cyclooxygenase-2 (COX-2) mRNA in colon cancer HT-29 cells. Methods: The total flavonoids of Cunninghamia lanceolata on human colon cancer HT-29 cells were observed by inverted microscope cell morphological changes, and set the total flavonoids high and low dose group and negative control group were 4 groups, using reverse transcription polymerization The expression level of COX-2 mRNA in cancer cells was detected by enzyme-linked-chain reaction (RT-PCR). The products of each amplified band were detected by automatic gel electrophoresis analysis system. Results: Compared with the negative control group, total flavonoids of Cuiyu grass can inhibit the expression of COX-2 at the mRNA level, and showed a dose-response relationship. CONCLUSION: The total flavonoids of Cuiyu grass can inhibit COX-2 at the mRNA level, thereby inhibiting the expression of COX-2 protein, which may be one of the mechanisms of anti-tumor effect of Cuiyu grass.
