目的 探讨急性全脑缺血损伤后大鼠海马神经细胞凋亡及相关蛋白Cpp3 2的基因表达、分布。方法 运用DNA梯形电泳、免疫印迹、免疫组化分别分析了全脑缺血后海马神经细胞损伤、Cpp3 2蛋白含量变化及其分布。结果 15min急性全脑缺血复灌 2 4h后 ,大鼠海马呈明显细胞凋亡特征 ,且天胱蛋白 (Cpp3 2 )基因被持续激活表达 ,尤其是锥体神经细胞表现最为明显。 结论 急性全脑缺血能诱导海马神经细胞凋亡及Cpp3 2蛋白基因表达 ,其中锥体神经细胞表现最显著
Objective To investigate the neuronal apoptosis and the gene expression and distribution of Cpp3 2 in rat hippocampus after acute global cerebral ischemia. Methods DNA ladder electrophoresis, Western blotting and immunohistochemistry were used to analyze the change and distribution of Cpp32 protein in hippocampal neurons after global cerebral ischemia. Results Apoptotic characteristics of rat hippocampus were observed after 15 min of reperfusion of acute global cerebral ischemia, and Cpp3 2 gene was activated continuously, especially in pyramidal neurons. Conclusion Acute global cerebral ischemia can induce the apoptosis of hippocampal neurons and the expression of Cpp3 2 protein. The expression of pyramidal neurons is the most significant
