目的分离及鉴定野生金线莲根内菌根真菌。方法采用平皿点种法进行金线莲菌根真菌的分离,提取菌株的总DNA,采用丝状真菌ITS通用引物对其rDNA相应区段进行PCR扩增,产物经纯化后测序。结果结合各菌株的形态特征和ITS区域基因序列特征,将A12菌株、5号菌株、2号菌株、6号菌株、A46菌株和A55菌株鉴定为丝核菌(Rhizoctonia sp.),其有性态为Thanatephorus sp.或Ceratobasidium sp.。结论通过对金线莲菌根真菌的分离鉴定,有望将其应用到金线莲及兰科其它濒危药用植物繁殖中去。
Objective To isolate and identify root mycorrhizal fungi of wild Anoectochilus. Methods The method of plate spotting was used to isolate Myrica rubra. The total DNA of the strain was extracted. The corresponding rDNA of the filamentous fungi ITS was amplified by PCR. The products were purified and sequenced. Results According to the morphological characteristics of each strain and ITS gene sequences, the strains A12, No.5, No.2, No.6 and A46 were identified as Rhizoctonia sp. Is Thanatephorus sp. Or Ceratobasidium sp. Conclusion Through the isolation and identification of mycorrhizal fungi, it is expected to be applied to the propagation of other endangered medicinal plants such as Anoectochilus and Orchidaceae.
