目的:建立HPLC法测定口服养阴通脑颗粒方后大鼠血清中阿魏酸浓度的方法。方法:健康SD大鼠ig养阴通脑颗粒水溶液后,制备含药大鼠血清。HPLC法测定大鼠含药血清中阿魏酸的含量。采用phenomenex GEMINI C18柱色谱柱,甲醇-3%乙酸(32∶68,v∶v)为流动相;流速1.0 mL.min-1;检测波长323 nm。结果:阿魏酸0.33~10.56μg.mL-1与峰面积线性关系良好(r=0.999 2),平均回收率为96.41%,RSD 2.36%(n=6);平均日内精密度RSD2.04%,平均日间精密度RSD5.04%,以信噪比S/N=3时测得的最低检测限为2.6 ng,血浆中的最低检测浓度为0.075μg.mL-1。结论:该方法简便,灵敏度高,稳定性及精密度较好,可用于大鼠血清中阿魏酸的含量测定。
Objective: To establish a HPLC method for the determination of ferulic acid in rat serum after oral administration of Yangyintongnao Granules. METHODS: After ig Yangyintongnao granule aqueous solution was prepared in healthy SD rats, drug-containing rat serum was prepared. HPLC determination of ferulic acid in rat drug-containing serum. A phenomenex GEMINI C18 column was used with methanol-3% acetic acid (32:68, v:v) as mobile phase; flow rate was 1.0 mL.min-1; detection wavelength was 323 nm. Results: The linear relationship between peak area and ferulic acid was 0.33~10.56μg.mL-1 (r=0.999 2). The average recovery was 96.41%, RSD was 2.36% (n=6), and the average intraday precision was 2.04%. The average precision of daytime RSD was 5.04%. The minimum detection limit was 2.6 ng when the signal-to-noise ratio S/N was 3, and the minimum detectable concentration in plasma was 0.075 μg.mL-1. Conclusion: The method is simple, sensitive, stable and accurate. It can be used for the determination of ferulic acid in rat serum.
