目的本研究旨在初探成都地区中老年人肠道双歧杆菌的种群分布的特点。方法收集成都地区2014年3~5月间45~74岁的正常健康中老年人的粪便标本60例,根据细菌的16Sr RNA基因序列设计双歧杆菌属及种的特异性引物,应用实时荧光定量PCR(Real-time quantitative PCR)反应对粪便标本中的双歧杆菌属及其人体最常见的8个种水平的进行定性定量分析。结果受试标本的双歧杆菌检出率是100%,平均检出4种以上双歧杆菌,检出3~6种的占70%以上;受试标本中的双歧杆菌数量平均值是8.2(以Log10为单位)、青春双歧杆菌(P<0.05)和角双歧杆菌(P<0.1)随年龄的增高,数量减少,其余6种双歧杆菌并没有随着年龄的增高出现统计学上的差异。婴儿双歧杆菌、短双歧杆菌的菌量随BMI的增高先增加后减少(P<0.05)。角双歧杆菌的数量(P<0.01)在不同性别表现不同。结论成都地区健康中老年人的肠道双歧杆菌数量显著地偏低,但是其菌种数却维持在较高的水平,和肠道双歧杆菌数量相比,肠道双歧杆菌数量菌种构成的多样性可能和中老年人健康有着更重要的关系。
Objective This study aimed to investigate the population distribution of bifidobacteria in the intestine of middle-aged and elderly people in Chengdu. Methods Sixty stool samples from 45 to 74 years of age from March to May, 2014 in Chengdu were collected. According to the 16S rRNA gene sequence of bacteria, specific primers of Bifidobacterium and species were designed and used for real-time fluorescence quantitative analysis Real-time quantitative PCR (PCR) was used to qualitatively and quantitatively analyze the most common 8 species of Bifidobacterium and their human bodies in stool specimens. Results The detection rate of bifidobacteria was 100% in the tested samples. Bifidobacteria were detected in more than 4 species on average, and more than 70% were detected among 3 to 6 species. The average number of bifidobacteria in the tested samples was 8.2 (Log10), Bifidobacterium adolescents (P <0.05), and Bifidobacterium hornbau (P <0.1) decreased with age, and the other 6 Bifidobacteria did not show statistical significance with age The difference. Bifidobacterium infantis and Bifidobacterium breve increased first and then decreased with the increase of BMI (P <0.05). The number of B. bifidum (P <0.01) differed by gender. Conclusion The number of intestinal Bifidobacteria in healthy middle-aged and elderly population in Chengdu is significantly lower than that in other regions. However, the number of Bifidobacterium is still relatively high in Chengdu. Compared with the number of Bifidobacterium in the intestine, The composition of the diversity may have a more important relationship with the health of the elderly.
