目的研究氯通道ClC-3和Cyclin D1在鼻咽癌细胞周期调控中的相互关系,探讨ClC-3参与细胞周期调控的可能机制。方法MTT法检测ClC-3反义寡核苷酸对鼻咽癌细胞增殖能力的影响,流式细胞仪测定细胞周期分布。用反义技术分别抑制细胞内ClC-3和Cyclin D1的表达,用RT-PCR检测目的基因的表达情况。结果40μg/ml ClC-3反义寡核苷酸能明显抑制鼻咽癌细胞的增殖,胞内ClC-3的表达下调对Cyclin D1表达没有影响,而Cyclin D1的表达下调则抑制鼻咽癌细胞ClC-3的表达。结论氯通道ClC-3参与了鼻咽癌细胞的周期调控,Cyclin D1可能位于该调控通路的上游。
Objective To study the relationship between ClC-3 and Cyclin D1 in the regulation of cell cycle of nasopharyngeal carcinoma and to explore the possible mechanism of ClC-3 involved in cell cycle regulation. Methods The effect of ClC-3 antisense oligonucleotide on the proliferation of nasopharyngeal carcinoma cells was detected by MTT assay. The cell cycle distribution was analyzed by flow cytometry. Antisense technology were used to inhibit the expression of ClC-3 and Cyclin D1, respectively. The expression of target gene was detected by RT-PCR. Results 40μg / ml ClC-3 antisense oligonucleotide could significantly inhibit the proliferation of nasopharyngeal carcinoma cells. The down-regulation of ClC-3 expression had no effect on the expression of Cyclin D1 while the down-regulation of Cyclin D1 inhibited the proliferation of nasopharyngeal carcinoma cells ClC-3 expression. Conclusion Chloride channel ClC-3 is involved in the cycle regulation of nasopharyngeal carcinoma cells. Cyclin D1 may be located upstream of this regulatory pathway.
