本文介绍高效液相色谱检测低氧时大鼠脑组织中腺苷酸,并将AMP、ADP、ATP分别定量。取大鼠脑组织0.5g以0.5mol/L高氯酸溶液处理后,用美国Waters-244型高效液相色谱,440型紫外检测器,波长254nm进行检测,用国产YWGC18不锈钢柱,流动相为甲醇:乙腈:IPRA:水=80:60:14:346(ml,V/V)。方法的回收率AMP、ADP、ATP分别为98.56%~100.48%,96.30%~103.7%,97.95%~99.56%;变异系数分别为4.08%、3.71%、3.68%。
This article describes the detection of adenylate in rat brain tissue by high performance liquid chromatography (HPLC), and quantifies AMP, ADP and ATP respectively. 0.5g rat brain tissue was treated with 0.5mol / L perchloric acid solution, and then detected by Waters-244 high performance liquid chromatograph, 440 UV detector and wavelength 254nm, with domestic YWGC18 stainless steel column Phase is methanol: acetonitrile: IPRA: water = 80: 60: 14: 346 (ml, V / V).
