目的研究桂枝芍药知母汤对Ⅱ型胶原诱导性关节炎大鼠的治疗作用,并探讨其作用机制。方法采用牛Ⅱ型胶原诱导的关节炎模型(Collagen-induced arthritis,CIA),灌胃给药21 d后观察大鼠体重变化、足肿胀度;采用ELISA法测定大鼠滑膜和血清中的白细胞介素4(IL-4)水平,小鼠巨噬细胞NO2-释放法测定大鼠滑膜和血清中干扰素γ(IFN-γ)活性。结果桂枝芍药知母汤能改善CIA大鼠体重变化,降低原发性和继发性足肿胀度,下调CIA大鼠滑膜和血清IFN-γ、IL-4水平。结论CIA大鼠滑膜中IFN-γ、IL-4水平均升高,血清中以IFN-γ升高为主。桂枝芍药知母汤抗风湿作用,可能通过下调CIA大鼠滑膜和血清IFN-γ、IL-4水平,从而调节Th1/Th2平衡。
Objective To study the therapeutic effects of Guizhi Shaoyao Zhimu Decoction on rats with type II collagen-induced arthritis and to explore its mechanism. Methods Collagen-induced arthritis (CIA) was induced by bovine collagen type II. Body weight changes and foot swelling were observed after intragastric administration for 21 days. White blood cells in synovium and serum of rats were measured by ELISA. Interleukin 4 (IL-4) levels and mouse macrophage NO2-release assay were used to measure interferon gamma (IFN-gamma) activity in synovial membranes and serum. Results Guizhi Shaoyao Zhimu Decoction can improve the weight change of CIA rats, reduce the degree of primary and secondary foot swelling, and down-regulate the levels of IFN-γ and IL-4 in synovium and serum of CIA rats. Conclusion The levels of IFN-γ and IL-4 in the synovium of CIA rats were increased, and the serum levels of IFN-γ were predominant. The anti-rheumatic effects of Guizhi Shaoyao Zhimu Decoction may regulate the Th1/Th2 balance by down-regulating the levels of synoviocytes and serum IFN-γ and IL-4 in CIA rats.
