本文探讨了样品处理方法对大鼠肌肉组织腺苷酸、乳酸和丙酮酸测定值的影响。结果表明:肌肉取出后迅速在液氮中夹冻并研磨成粉,在4℃用玻璃匀浆器匀浆能较完全地破碎细胞膜,得到满意的测定值。再经超声波处理,线粒体等亚细胞结构完全破碎,ATP和总腺苷酸(TAN)的测定值增加20%,乳酸和丙酮酸变化不大。超声波处理在1min以内AMP和ADP变化不明显,时间过长则随时间而升高。
This article explored the effect of sample handling on the determination of adenosine, lactate and pyruvate in rat muscle tissue. The results showed that the muscles were quickly frozen in liquid nitrogen and ground into powder. The homogenate was homogenized with glass homogenizer at 4 ° C to completely break the cell membrane, and satisfactory results were obtained. After sonication, mitochondria and other subcellular structures were completely broken. The measured values of ATP and total adenosine monophosphate (TAN) increased by 20%, and the changes of lactate and pyruvate did not change much.
