目的探讨生长分化因子-15(GDF-15)在子痫前期患者胎盘滋养细胞中的表达,探讨其在子痫前期发病机制中的作用。方法选择2009年12月至2010年10月在青岛市市立医院住院分娩的子痫前期孕妇140例,其中早发型轻度子痫前期孕妇35例(早发轻度组),晚发型轻度子痫前期孕妇35例(晚发轻度组),早发型重度子痫前期孕妇35例(早发重度组),晚发型重度子痫前期孕妇35例(晚发重度组);另选同期健康孕妇35例为对照组。采用RT-PCR检测胎盘滋养细胞中GDF-15 mRNA的表达量;采用免疫组化方法检测胎盘滋养细胞中GDF-15蛋白的表达。结果 RT-PCR显示子痫前期各组胎盘滋养细胞中GDF-15mRNA表达水平均高于对照组,差异有统计学意义(P<0.05);但子痫前期各组间分别比较,差异无统计学意义(P>0.05)。免疫组化显示GDF-15表达于胎盘滋养细胞的细胞浆和细胞外基质中,并在各组胎盘滋养细胞中均表达。子痫前期各组胎盘滋养细胞中GDF-15蛋白表达水平均高于对照组组,差异有统计学意义(P<0.05);但子痫前期各组间分别比较,差异无统计学意义(P>0.05)。结论 GDF-15在子痫前期胎盘滋养细胞中表达升高,GDF-15的表达水平变化可能与子痫前期发病有关。
Objective To investigate the expression of GDF-15 in placental trophoblast cells of preeclampsia and to explore the role of GDF-15 in the pathogenesis of preeclampsia. Methods Forty pregnant women with preeclampsia who were hospitalized in Qingdao Municipal Hospital from December 2009 to October 2010 were selected. Among them, 35 were premature pregnant women with mild preeclampsia (early-onset mild group), late-onset mild child 35 pregnant women with preeclampsia (mild late onset group), 35 pregnant women with early-onset severe preeclampsia (early-onset severe group), 35 late pregnant women with severe preeclampsia (severe late-onset group) 35 cases as control group. The expression of GDF-15 mRNA in placental trophoblast cells was detected by RT-PCR. The expression of GDF-15 protein in placental trophoblast cells was detected by immunohistochemistry. Results RT-PCR showed that the expression level of GDF-15 mRNA in placental trophoblast cells in preeclampsia group was significantly higher than that in control group (P <0.05), but there was no significant difference between preeclampsia groups Significance (P> 0.05). Immunohistochemistry showed that GDF-15 was expressed in the cytoplasm and extracellular matrix of placental trophoblasts and expressed in all groups of placental trophoblasts. The expression of GDF-15 protein in placental trophoblast cells in preeclampsia group was higher than that in control group (P <0.05), but there was no significant difference between preeclampsia groups (P > 0.05). Conclusion The expression of GDF-15 in preeclampsia placental trophoblast cells is increased, and the change of GDF-15 expression may be related to the pathogenesis of preeclampsia.
