目的观察不同剂量的黄芪多糖(APS)对糖尿病大鼠胰腺Kir6.2表达的干预作用。方法建立Wistar大鼠糖尿病模型,随机分为模型组、对照组、APS低剂量干预组和APS高剂量干预组,进行糖耐量实验(OGTT),检测胰腺Kir6.2mRNA和蛋白表达变化。结果模型组OGTT30min、60min、120min和180min血糖值显著高于对照组(P<0.05),而APS高剂量干预组的60min和120min血糖值较模型组显著降低(P<0.05);与对照组比较,模型组胰腺Kir6.2mRNA和蛋白表达水平显著下降(P<0.05),APS低剂量干预组和APS高剂量干预组Kir6.2的mRNA和蛋白表达水平较模型组有不同程度上调(P<0.05)。结论黄芪多糖对糖尿病大鼠胰腺Kir6.2表达有一定的上调作用,可能是其发挥降糖作用的机制之一。
Objective To observe the effects of different doses of astragalus polysaccharide (APS) on the expression of Kir6.2 in the pancreas of diabetic rats. Methods The diabetic model of Wistar rats was established and randomly divided into model group, control group, low dose APS intervention group and high dose APS intervention group. The glucose tolerance test (OGTT) was performed to detect the expression of Kir6.2 mRNA and protein in pancreas. Results The blood glucose levels of OGTT at 30min, 60min, 120min and 180min in model group were significantly higher than those in control group (P <0.05), while those in high-dose APS group at 60min and 120min were significantly lower than those in model group (P <0.05) (P <0.05). The levels of Kir6.2 mRNA and protein in pancreas in model group were significantly lower than those in model group (P <0.05) ). Conclusion Astragalus polysaccharide can up-regulate the expression of Kir6.2 in the pancreas of diabetic rats, which may be one of the mechanisms by which astragalus polysaccharide can exert hypoglycemic effect.
