沉默信息调节因子2相关酶1(silent information regulator 2-related enzymes 1,Sirtuin 1,SIRT1)是一种烟酰胺腺嘌呤二核苷酸(nicotinamide adenine dinucletide,NAD)依赖性的去乙酰化酶,通过对组蛋白、非组蛋白及其他转录因子的调节参与了体内许多生理功能的调节,包括众多基因转录、能量代谢、氧化应激以及细胞衰老过程等。最近研究证实,SIRT1在多种病理过程中,通过调节内皮源型一氧化氮合酶(endothelial nitric oxide syntheses,eNOS)、p53、叉头转录因子(forkhead box class O,FOXO)、核转录因子-κB(nuclear factor kappa B,NF-κB)等的活性表现出血管内皮及神经功能保护作用。因此,对于以内皮功能障碍为主要病因的疾病特别是勃起功能障碍(erec-tile dysfunction,ED),SIRT1可能是其潜在的治疗靶点之一。
SIRT1 (Sirtuin 1, SIRT1) is a nicotinamide adenine dinucletide (NAD) -dependent sirtuin that is expressed by Regulation of histones, non-histones and other transcription factors is involved in the regulation of many physiological functions in the body, including numerous gene transcription, energy metabolism, oxidative stress and cellular senescence. Recent studies have demonstrated that SIRT1 regulates endothelium-derived endothelial nitric oxide synthase (eNOS), p53, forkhead box class O (FOXO), and nuclear transcription factor- κB (nuclear factor kappa B, NF-κB) activity showed vascular endothelial and neuroprotective effects. Therefore, SIRT1 may be one of its potential therapeutic targets for diseases with endothelial dysfunction as the main cause, especially erec-tile dysfunction (ED).
