未分离的人骨髓在冰冻和解冻后,有许多成熟粒系细胞破碎后释放出 DNA 和溶酶体中的酶,它们引起细胞凝块导致细胞数的减少和细胞活力的降低。因而作者研究了在冰冻前即去除成熟粒细胞和红细胞的各种方法。从病人多部位穿刺取得骨髓,用不含防腐剂的肝素抗凝。500~600毫升骨髓与150~200毫升 TC199混合,通过粗的(0.307mm)和细的(0.201mm)筛网,得到单一细胞悬液,其中约有2×10~(10)有核细胞。骨髓在分离前用 Hanks 液按1∶1稀释是一个重要步骤,否则会影响回收率。
Non-isolated human bone marrow After freezing and thawing, many mature granulocytes break down and release enzymes in DNA and lysosomes, causing them to cause cell clots to decrease in number and cellular viability. Thus the authors studied various ways to remove mature granulocytes and erythrocytes before freezing. Bone marrow was obtained from multiple parts of the patient and anticoagulated with a preservative-free heparin. 500 to 600 ml of bone marrow is mixed with 150 to 200 ml of TC199 and passed through a coarse (0.307 mm) and fine (0.201 mm) screen to obtain a single cell suspension of about 2 × 10 10 nucleated cells. It is an important step to dilute the bone marrow with 1: 1 Hanks solution before isolation, otherwise the recovery will be affected.
