目的:建立乌拉儿甘草总多糖最佳提取工艺。方法:采用水提醇沉法,以总多糖提取率为指标,通过单因素与正交试验L9(34)相结合,考察提取粒度、温度、时间、料液比、乙醇浓度和提取次数对甘草多糖提取工艺的影响。结果:甘草多糖提取脱脂条件为粉碎粒度80目,乙醇浓度80%,料液比1∶5(g/v),时间40 min/次,脱脂1次;提取条件为提取溶剂蒸馏水,料液比1∶20(g/v),提取温度75℃,提取时间30 min/次,提取5次;多糖提取后以80%乙醇4℃醇沉24h。结论:该工艺简便、合理,多糖提取率高。
Objective: To establish the optimum extraction process of total polysaccharide of Urath licorice. Methods: The water extraction and alcohol precipitation method was used to determine the extraction rate of total polysaccharides. The effects of extraction granularity, temperature, time, ratio of solid to liquid, ethanol concentration and extraction times were studied by single factor and orthogonal experiment L9 (34) Effect of polysaccharide extraction process. Results: The extraction and degreasing conditions of glycyrrhiza polysaccharides were 80 mesh size, 80% ethanol concentration, 1: 5 (g / v) solid-to-liquid ratio, 40 min time and 1 time degreasing. The extraction conditions were extraction solvent distilled water, 1:20 (g / v), extraction temperature 75 ℃, extraction time 30 min / time, extraction 5 times; polysaccharide extracted with 80% ethanol 4 ℃ alcohol precipitation 24h. Conclusion: The process is simple and reasonable, polysaccharide extraction rate is high.
