目的:探讨桃仁对单侧输尿管结扎诱导的肾间质纤维化实验大鼠肾小管上皮细胞转分化的影响。方法:采用单侧输尿管结扎(UUO)肾间质纤维化模型。将大鼠随机分为4组,正常组、模型组ig给2 mL自来水,蒙诺组ig予10 mg·kg-1和桃仁组ig予6 g·kg-1释成冲剂2 mL,每日1次。术后14 d处死大鼠,观察肾组织病理改变,并应用免疫组织化学方法检测肾组织上皮细胞钙黏蛋白(E-cadherin),α-平滑肌激动蛋白(α-SMA)和纤维连接蛋白(FN)的表达。结果:与正常组相比,模型组肾小管上皮细胞E-cadherin有较弱表达,肾间质α-SMA和FN有较强的表达(P<0.05);与模型组相比,桃仁组纤维化程度减轻,且α-SMA和FN的阳性表达较模型组显著减弱,E-cadherin表达较强,有统计学差异(P<0.05),桃仁组与蒙诺组比无统计学差异少。结论:桃仁可能通过增强E-cadherin、减弱α-SMA和FN的表达改善肾小管上皮细胞转分化,而减缓肾间质纤维化病程进展。
Objective: To investigate the effect of peach kernel on renal tubular epithelial-myofibroblast transdifferentiation induced by unilateral ureteral ligation in rats with renal interstitial fibrosis. Methods: Unilateral ureteral obstruction (UUO) model of renal interstitial fibrosis was used. The rats were randomly divided into 4 groups, normal group, model group ig to 2 mL of tap water, Monopril group ig to 10 mg · kg-1 and peach kernel group ig to 6 g · kg-1 release granules 2 mL, daily 1 time. The rats were sacrificed 14 days after operation to observe the pathological changes of renal tissue. Immunohistochemistry was used to detect the expression of E-cadherin, α-smooth muscle actin (α-SMA) and fibronectin (FN )expression. Results: Compared with the normal group, the expression of E-cadherin in renal tubular epithelial cells of the model group was weaker than that of the normal group (P <0.05). Compared with the model group, (P <0.05). There was no significant difference in the expression of α-SMA and FN between the peach kernel group and the Monel group. The positive expression of α-SMA and FN was significantly weaker than that of the model group and the expression of E-cadherin was stronger. Conclusion: Peach kernel may improve the renal tubular epithelial cell transdifferentiation and decrease the progression of renal interstitial fibrosis by enhancing the expression of E-cadherin, attenuating the expression of α-SMA and FN.
