目的研究血管紧张素Ⅱ(AngⅡ)、他汀类药物及普罗布考干预后对骨形态发生蛋白2(BMP-2)表达的影响,初步探讨BMP-2表达的可能调节机制。方法分组培养人脐静脉内皮细胞,加入不同浓度的AngⅡ,并分别用阿伐他汀、普罗布考干预;用RT-PCR法检测BMP-2的表达,ELISA法测定细胞上清液中BMP-2水平,比色法检测丙二醛(MDA)和超氧歧化酶(SOD)的水平。结果1)一定浓度的AngⅡ可以增加BMP-2和核因子-κΒ蛋白65(NF-κΒ-p65)的表达。2)阿伐他汀和普罗布考干预后,BMP-2、NF-κΒ-p65的表达下降,过氧化物酶体增殖物激活受体γ(PPARγ)表达加强,同时可逆转由AngⅡ干预导致的MDA水平升高、SOD水平下降的趋势。结论AngⅡ增加BMP-2的表达,可能与增加NF-κΒ-p65的表达有关,而阿伐他汀和普罗布考降低BMP-2的表达可能与下调NF-κΒ-p65的表达、激活PPARγ的表达有关。
Objective To investigate the effect of angiotensin Ⅱ (AngⅡ), statins and probucol on the expression of bone morphogenetic protein 2 (BMP-2), and to explore the possible regulatory mechanism of BMP-2. Methods Human umbilical vein endothelial cells were cultured in different groups with Ang Ⅱ supplemented with atorvastatin and probucol respectively. The expression of BMP-2 was detected by RT-PCR and the expression of BMP-2 The levels of malondialdehyde (MDA) and superoxide dismutase (SOD) were measured by colorimetry. Results 1) A certain concentration of AngⅡ could increase the expression of BMP-2 and NF-κB protein 65 (NF-κB-p65). 2) After intervention with atorvastatin and probucol, the expression of BMP-2 and NF-κB-p65 decreased and the expression of peroxisome proliferator activated receptorγ (PPARγ) MDA levels, SOD levels decreased trend. Conclusion AngⅡ increased BMP-2 expression may be related to the increase of NF-κB-p65 expression, while atorvastatin and probucol reduce BMP-2 expression may be related to down-regulation of NF-κΒ-p65 expression, activation of PPARγ expression related.
