目的:探讨复方“别敏”中黄芩苷、淫羊藿苷与黄芪甲苷3种成分合用的优化配伍比例。方法:采用卵清蛋白腹腔注射建立变应性鼻炎小鼠模型,以脾淋巴细胞增殖反应为指标,利用四唑氮氢氧化物法确定黄芩苷、淫羊藿苷、黄芪甲苷各成分的有效剂量范围;选用U1*0(108)表组方设计所得的3种成分的10种组合进行干预,考察其对脾淋巴细胞增殖的抑制效应,经逐步回归统计分析,获得3种成分合用的最佳配比;通过淋巴细胞增殖反应和对淋巴细胞培养上清液中细胞因子含量检测,验证3种成分的最佳配伍效应是否优于各单一成分。结果:2~10μmol/L黄芩苷、2~10μmol/L淫羊藿苷和1~10μmol/L黄芪甲苷明显抑制致敏小鼠脾淋巴细胞增殖;当黄芩苷、淫羊藿苷、黄芪甲苷以1:2.14:2.65比例配伍时,抑制细胞增殖的效应最明显;进一步的验证研究证实,该配伍对ConA刺激的淋巴细胞增殖的抑制效果及对淋巴细胞上清液中白细胞介素4(interleukin-4,IL-4)、IL-5和γ干扰素(interferon-gamma,IFN-γ)的调节作用明显优于黄芩苷、淫羊藿苷、黄芪甲苷单用的效果(P<0.05,P<0.01)。结论:复方“别敏”中主要有效成分黄芩苷、淫羊藿苷、黄芪甲苷按优化比例合用可最有效地抑制致敏小鼠脾淋巴细胞增殖,调节因变态反应引起的Th1、Th2型细胞因子失衡。
OBJECTIVE: To investigate the optimal ratio of combination of baicalin, icariin and astragaloside in compound prescription “Bie Min”. Methods: A mouse model of allergic rhinitis was established by intraperitoneal injection of ovalbumin. The spleen lymphocyte proliferative response was used as an index to determine the effects of various components of baicalin, icariin and astragaloside IV Dose range; the use of U1 * 0 (108) table design of the three components of the 10 combinations of interventions to investigate its inhibitory effect on splenic lymphocyte proliferation, by stepwise regression analysis, access to the three components combined with the most Good ratio; through lymphocyte proliferation reaction and detection of cytokines in lymphocyte culture supernatant to verify the best compatibility effect of the three components is superior to each single component. Results: 2 ~ 10μmol / L Baicalin, 2 ~ 10μmol / L icariin and 1 ~ 10μmol / L Astragaloside significantly inhibited the proliferation of splenic lymphocytes in sensitized mice. When Baicalin, Icariin, Astragalus A Glycosides in 1: 2.14: 2.65 ratio compatibility inhibition of cell proliferation the most obvious effect; further validation studies have confirmed that the compatibility of ConA-stimulated lymphocyte proliferation inhibitory effect on lymphocyte supernatant interleukin 4 ( interleukin-4, IL-4, IL-5 and interferon-gamma (IFN-γ) were significantly better than those of baicalin, icariin and astragaloside alone , P <0.01). Conclusion: Baicalin, icariin and astragaloside, the main active ingredient in compound Bianmin, can inhibit the proliferation of splenic lymphocytes in sensitized mice most effectively and regulate the expression of Th1, Th2-type cytokines imbalance.
