目的探讨KCNJ1基因rs675759、rs675388和rs2846679位点基因型与浙江省宁波地区汉族人群血脂水平和血脂异常发病风险的相关性。方法以浙江省宁波市鄞州区作为研究现场,选取年龄≥40岁的汉族常住居民2 330例作为研究对象。流行病学调查收集样本人群一般信息、生活行为方式等资料,采集血液样本。聚合酶链反应-连接酶检测技术检测KCNJ1 rs675759、rs675388和rs2846679位点基因型,Hardy-Weinberg平衡检验样本的遗传平衡状态。采用多重线性回归分析各位点不同基因型对血浆总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)血脂水平的影响。结果 KCNJ1基因的3个位点分型成功率均>99%,Hardy-Weinberg平衡检验结果显示均P>0.05。rs675759位点每个G等位基因使HDL-C减少0.12 mmol/L;rs675388位点每个T等位基因使TC水平增加0.14 mmol/L,HDL-C增加0.12 mmol/L;rs2846679位点每个A等位基因使TC降低0.09 mmol/L,TG下降0.08 mmol/L。结论在本研究的样本人群中,KCNJ1基因多态性与TC、TG和HDL-C水平相关。
Objective To investigate the association between the genotypes of rs675759, rs675388 and rs2846679 in KCNJ1 gene and the risk of developing dyslipidemia in Han nationality in Ningbo, Zhejiang Province. Methods Taking Yinzhou District of Ningbo City in Zhejiang Province as a research site, 2 330 Han residents aged 40 years or older were selected as the research object. Epidemiological survey collected samples of the general population information, lifestyle behavior and other data collection of blood samples. Polymerase chain reaction-ligase assay was used to detect the genetic equilibrium of KCNJ1 rs675759, rs675388 and rs2846679 genotypes and Hardy-Weinberg equilibrium test samples. Multiple linear regression was used to analyze the effect of different genotypes on plasma total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL-C) and high density lipoprotein cholesterol (HDL-C) . Results The success rate of KCNJ1 gene typing was> 99% at three loci, and the Hardy-Weinberg equilibrium test showed that both P> 0.05. Each allele of rs675759 reduced HDL-C by 0.12 mmol / L; each T allele of rs675388 increased TC level by 0.14 mmol / L and HDL-C by 0.12 mmol / L; rs2846679 locus per The A allele decreased TC by 0.09 mmol / L and TG by 0.08 mmol / L. Conclusions In our sample population, KCNJ1 gene polymorphisms were associated with TC, TG and HDL-C levels.
